ABSTRACT

Abstract Objectives: To evaluate the acquisition of surgical skills by otolaryngology residents and established the minimum number of dissections of a lamb's model to be performed before practicing on human patients. Methods: Nineteen second-year otolaryngology residents performed ten dissections each, five on each nasal cavity, always practicing the same three surgical procedures on the lamb model. Each student's training lasted 2-months, and the entire training intervention lasted 4-years, over four generations of residents. All dissections were recorded and were selected at random for examination by two independent otolaryngology surgeons, who were otherwise not involved in the research. Assessment of the 190 dissections used an instrument validated for surgical training of medical residents. Results: To a 1% significance level, statistical analysis revealed increased performance and satisfactory results were observed after the sixth dissection. Furthermore, after the eighth dissection, skill acquisition was relevant and sustained. Conclusion: Training in endoscopic nasal surgery on a lamb's head model improves surgical skills and handling of surgical instruments. Our results showed the relevance of the lamb model for training in otolaryngology surgery, impacting on patient safety. Level of evidence: 2.

ABSTRACT

Chronic atrophic gastritis (CAG) and gastric intestinal metaplasia (GIM) significantly increase the risk of gastric cancer. Blocking the development of CAG and GIM would help to decrease the incidence of gastric cancer. It was revealed that eradication of Helicobacter pylori could reverse gastric mucosa atrophy. Recent studies reported that GIM could be reversed to a certain extent. Clinical studies demonstrated that Lamb′s tripe extract and vitamin B12 capsule exhibited significant reversal effects on GIM. The present study systemically reviewed the diagnosis and treatment of CAG and clinical application of Lamb′s tripe extract and vitamin B12 capsule, and established the specialist instruction that would guide the reversal treatment of CAG and GIM.

ABSTRACT

Abstract Congenital malformations are abnormalities of structure, formation or function observable at birth that causes great economic losses in livestock industry. In this study, we describe the incidence of the main congenital anomalies encountered in the last year (June 2020-June 2021) in ruminants from Elazig region according to species, breed, age and sex, number of pregnancies, rectal examination, and method of conception employed. A total of 79 congenital anomalies encountered in 73 animals during this study were evaluated. Of 73 animals most of congenital anomalies were calves (84.93%) and males (68.49%). It was determined that the most common type of congenital anomaly in calves was hernia umbilicalis (21.51%) while in lambs and goat kids was atresia ani. Most of these anomalies were formed in the abdominal wall and organs of gastrointestinal system (46.84%). Congenital anomalies in calves and lambs were more frequently when they came from the first pregnancy of their dams, while in goat kids when their dam had the second pregnancy. Most of small ruminant that had congenital anomalies came from dams with multiple gestation. Rectal examinations performed in cattle during the early stages of gestation was probably one of a predisposing factor for the formation of congenital malformations in this specie. More studies that include the participation of diagnostic laboratories are needed to improve the characterization of congenital anomalies that contribute to the implementation of preventive and control measures in the herds of these species in our region.


Resumen Las malformaciones congénitas son anomalías de estructura, formación o función observables al nacer que provocan grandes pérdidas económicas en la industria ganadera.. En este estudio, se investigó la incidencia de las principales anomalías congénitas encontradas en el último año (junio de 2020-junio de 2021) en rumiantes en la región de Elazig. según especie, raza, edad, sexo, número de preñeces, examen rectal y método de concepción empleado . Se evaluaron un total de 79 anomalías congénitas encontradas en 73 animales . De 73 animales con anomalías congénitas, la mayoría eran terneros (84, 93%) y machos (68,49%). Se determinó que el tipo de anomalía más común en los terneros fue la hernia umbilical (21,51%) mientras que en corderos y cabritos fue la atresia anal. La mayoría de estas anomalías se formaban principalmente en los órganos de la pared abdominal y órganos del sistema gastrointestinal (46,84%). . Las anomalías congénitas en terneros y corderos eran más frecuentes cuando provenían de las madres de primera preñez , mientras que en los cabritos cuando las madres tenían su segunda preñez La mayoría de los rumiantes menores con anormalidades congénitas provenían de madres con gestaciones múltiples. El examen rectal realizado en las vacas durante los primeros estadios de la gestación, fue probablemente uno de los factores predisponentes para la formación de malformaciones congénitas en esta especie. Se requieren más estudios que incluyan la participación de laboratorios de diagnóstico para mejorar la caracterización de las anormalidades congénitas y contribuir a la implementación de medidas de prevención y control en los rodeos de estas especies en nuestra región.

Abstract Background: We hypothesized that a diet with old man saltbush hay used as an alternative source of nutrients could partially replace the concentrate in the feeding of feedlot lambs. Objective: This study evaluated the behavior and performance of lambs fed diets containing increasing levels of old man saltbush hay plus concentrate. Methods: Twenty-four castrated Santa Inês lambs at approximately eight months of age (22 ± 1.97 kg) were confined in a randomized complete design and fed diets containing 30, 40, 50, and 60% (dry matter: DM) of old man saltbush hay. Results: The intake of DM and neutral detergent fiber was not affected (p>0.05) by the level of old man saltbush hay. Intake of mineral salt decreased (p<0.01) as the proportion of old man saltbush in the diet increased. Weight gain (kg) and feed efficiency worsened (p<0.05) with 40% inclusion of old man salt bush hay. The time spent on feeding and chewing increased (p<0.05), while idle time was reduced (p<0.01) with dietary inclusion of old man saltbush. Feed efficiency decreased (p<0.05) with the inclusion of old man saltbush. Conclusion: Dietary inclusion of old man saltbush affects the feeding behavior and performance of lambs in feedlot.


Resumen Antecedentes: Nuestra hipótesis consistió en que una dieta con heno de hierba-sal como fuente alternativa de nutrientes puede sustituir parcialmente el concentrado en la alimentación de corderos confinados. Objetivo: Evaluar el comportamiento ingestivo de corderos alimentados con dietas conteniendo niveles incrementales de heno de hierba-sal asociado al concentrado. Métodos: Veinticuatro corderos Santa Inés, castrados, con aproximadamente ocho meses de edad, peso corporal de 22 ± 1,97 kg, confinados, fueron distribuidos en un arreglo experimental completamente aleatorizado y alimentados con dietas conteniendo 30, 40, 50 y 60% (materia seca: MS) de heno de hierba-sal. Resultados: El consumo de MS y fibra detergente neutra no se vió afectado (p>0,05) por los diferentes niveles de inclusión de hierba-sal. La ingestión de sal mineral se redujo (p<0,01) al incrementar la hierba-sal en la dieta. La ganancia de peso (kg) y la eficiencia alimenticia empeoraron (p<0,05) con la inclusión de 40% de heno de hierba-sal. El tiempo empleado para las actividades de alimentación y masticación aumentó (p<0,05), mientras que el de ocio se redujo (p<0,01) con la inclusión de hierba-sal. La eficiencia alimenticia de la materia seca disminuyó (p<0,05) a medida que aumentó el nivel de inclusión de heno de hierba-sal. Conclusión: La inclusión de heno de hierba-sal en la dieta afecta el comportamiento ingestivo y el rendimiento de corderos confinados.


Resumo Antecedentes: A hipótese foi que uma dieta com o feno de erva-sal como fonte alternativa de nutrientes poderia substituir parcialmente o concentrado na alimentação de cordeiros confinados. Objetivo: Este estudo foi desenvolvido para avaliar o comportamento ingestivo de cordeiros alimentados com dietas contendo feno de erva-sal associado com o concentrado. Métodos: Vinte e quatro cordeiros Santa Inês, castrados, com aproximadamente oito meses de idade, peso corporal de 22 ± 1,97 kg, confinados, foram distribuídos em um delineamento experimental inteiramente casualizado e alimentados com dietas contendo 30, 40, 50 e 60% (matéria seca (MS)) de feno de erva-sal. Resultados: O consumo de matéria seca e fibra em detergente neutro não foram comprometidos (p>0,05) pelos diferentes níveis de erva-sal. A ingestão de sal mineral reduziu (p<0,01) com o aumento de erva sal nas dietas. O ganho de peso e a eficiência alimentar pioraram (p<0,05) a partir da inclusão de 40% de feno de erva-sal na dieta dos cordeiros. O tempo despendido para as atividades de alimentação e mastigação aumentaram (p<0,05), enquanto que o de ócio reduziu (p<0,01) com a inclusão da erva sal nas dietas. A eficiência de alimentação da matéria seca diminuiu (p<0,05) à medida que se incluiu o feno de erva-sal. Conclusão: A inclusão de feno de erva-sal na dieta afeta o comportamento ingestivo eo desempenho de cordeiros confinados.

Abstract Background: The use of pequi by-product as animal feed is an appealing alternative to reduce overall production costs. However, little is known regarding its effects on animal performance as well as the ideal inclusion level in the diet. Objective: To evaluate performance, intake, and digestibility of confined lambs receiving diets containing different levels of pequi by- product as a partial substitute for corn silage. Methods: The experiment was conducted in Curvelo, Brazil, using twenty-five crossbred Santa Inês male lambs (17.83 ± 1.73 kg) aged six months for 92 days. Increasing levels of pequi by-product (0, 6, 12, 18, and 24%) replacing corn silage were evaluated in a completely randomized design. Analysis of variance and regression were performed, with 5% significance level. Results: Inclusion of pequi by-product did not influence (p>0.05) average daily gain, total weight gain, or feed conversion ratio. No effect (p>0.05) was also observed on intake of dry matter, organic matter, ether extract, crude protein, neutral detergent fiber, total carbohydrates, and total digestible nutrients. However, a linear decrease was observed (p<0.05) in the apparent digestibility of dry matter, organic matter, ether extract, crude protein, neutral detergent fiber, total carbohydrates, and total digestible nutrients with increasing inclusion of pequi by-product. Conclusion: Pequi by-product is a potential substitute for corn silage in lamb diets because it does not influence animal performance and feed intake. However, digestibility decreases with increasing dietary inclusion levels of pequi by-product.


Resumen Antecedentes: la inclusion de subproductos de pequi en alimentación animal es una estrategia atractiva para reducir los costos de producción. Sin embargo, todavía se sabe poco sobre el efecto de este subproducto en el rendimiento de los animales, así como su nivel ideal de inclusión en la dieta. Objetivo: evaluar el rendimiento, consumo y digestibilidad en corderos confinados recibiendo dietas con subproducto de pequi en reemplazo parcial del ensilado de maíz. Métodos: el experimento se realizó en Curvelo, Brasil, utilizando 25 corderos machos cruzados de raza Santa Inês (17,83 ± 1,73 kg) y 6 meses de edad, durante 92 días. Se evaluaron niveles crecientes del subproducto de pequi (0, 6, 12, 18 y 24%) para sustituir el ensilado de maíz siguiendo un diseño completamente aleatorio. Los análisis de varianza y regresión se llevaron a cabo usando un nivel de significancia del 5%. Resultados: la inclusión del subproducto de pequi no influyó (p>0,05) en la ganancia de peso diaria, ganancia de peso total, ni conversión alimenticia. No se observó efecto (p>0,05) en la ingesta de materia seca, materia orgánica, extracto etéreo, proteína bruta, fibra detergente neutra, carbohidratos totales y nutrientes digestibles totales. Sin embargo, se observó una reducción lineal (p<0,05) en la digestibilidad aparente de materia seca, materia orgánica, extracto etéreo, proteína bruta, fibra detergente neutra, carbohidratos totales y nutrientes digestibles totales con el aumento del nivel de inclusión del subproducto. Conclusión: el subproducto de pequi puede utilizarse como sustituto parcial del ensilado de maíz, ya que no afecta la ganancia de peso ni la ingesta de nutrientes. Sin embargo, la digestibilidad disminuye al aumentar el nivel de inclusión del subproducto en la dieta.

Objetivo: avaliar o desempenho, consumo e digestibilidade em cordeiros confinados recebendo dietas contendo subproduto de pequi em substituição à silagem de milho. Métodos: o experimento foi conduzido em Curvelo, Brasil, utilizando 25 cordeiros machos cruzados da raça Santa Inês (17,83 ± 1,73 kg) com 6 meses de idade durante 92 dias. Níveis crescentes do subproduto de pequi (0, 6, 12, 18 e 24%) foram avaliados em substituição à silagem de milho em um delineamento inteiramente casualizado. A análise de variância e regressão foram conduzidas adotando 5% como nível de significância. Resultados: a inclusão do subproduto de pequi não influenciou (p>0,05) o ganho de peso médio diário, o ganho de peso total e a conversão alimentar. Conclusão: o subproduto de pequi pode ser utilizado como substituto parcial à silagem de milho uma vez que não influenciou o desempenho animal ou o consumo de nutrientes. No entanto, a digestibilidade diminuiu com o aumento do nível de inclusão do subproduto na dieta.

ABSTRACT

Objective:To study the material basis of the drug effect of the raw material of lamb′s tripe and vitamin B12 capsule.Methods:Rennet and pepsin were extracted with 0.9% sodium chloride and were purified by saturated ammonium sulfate precipitation, diethylaminoethyl-cellulose 52 and high pressure chromatography (HPLC) chromatography. Relative molecular weight was determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) electrophoresis and liquid chromatography-triple quadrupole mass spectrometer. The amino acid composition and antioxidant activity of raw materials were analyzed with HPLC. The raw materials were completely extracted with water, phosphate buffer and sodium bicarbonate in turn, and in vitro 1, 1-diphenyl-2-picrylhydrazyl radil 2, 2-diphenyl-1-(2, 4, 6-trinitrophenyl)hydrazyl (DPPH) and 2, 2′-azinobis-(3-ethylbenzthiazoline-6-sulphonate) (ABTS) radical activity were measured. Glycoprotein from raw material was extracted with hot water and determined its growth-promoting activity gaginst Bifidobacterium adolescentis , Lactobacillus delbrueckii subsp. Bulgaricus and Enterococcus faecalis was measured. The composition of amino acid and monosaccharide were analyzed by HPLC and gas chromatography-mass spectrometry, respectively. Results:The enzyme activity of two purified rennet F6-2 and F7-2 which had different ionic strengths and pepsin F7-1 were 27 557.10, 17 532.60 and 17 728.15 U/g, respectively. The results of SDS-PAGE indicated that the molecular weights of three enzymes were similar, ranging from 35 000 to 40 000. The raw material contained 16 kinds of amino acids, of which hydrophobic amino acids accounted for 33.03% of the total amino acid content. When the sample concentration was 5 mg/mL, ABTS free radical scavenging activity of the three extracts was (37.80±0.45)%, (23.20±0.78)% and (62.80±0.74)%; DPPH free radical scavenging activity was (57.87±0.55)%, (5.03±0.25)% and (26.67±3.10)%, respectively. Glycoprotein extracts had promoted the growth of Bifidobacterium adolescentis, Lactobacillus delbrueckii subsp. Bulgaricus and Enterococcus faecalis, and there was statistically significant difference in the promotion of Lactobacillus delbrueckii subsp. Bulgaricus and Enterococcus faecalis ( P<0.05). The protein chain of glycoprotein was composed of 15 amino acids and the polysaccharide chain was composed of two monosaccharides, glucose and lactose. Conclusions:Two rennet and one pepsin are isolated and analyzed from raw material of lamb abomasum by various chromatographic methods. The raw material is rich in antioxidant active ingredients. The glycoprotein components of lamb abomasum has the activity of promoting the growth of probiotics.

ABSTRACT

Objective:To explore the therapeutic effects of lamb′s tripe extract and vitamin B12 (hereinafter referred to as lamb′s tripe) capsule on precancerous lesions of gastric mucosa in rats.Methods:Thirty-two rats of the 42 Wistar rats (model group) were selected for modelling, and in model group six rats died due to gavage, 10 rats were sacrificed for observing the results of modeling, and the left 16 rats were divided into administration group (eight rats) and non-administration group (eight rats). After modeling, five of the 10 rats without modelling treatment were selected as the normal control of the model group, the other five (negative control group) rats were included in drug intervention experiment. The drug intervention program was as follows: in administration group, rats were gavaged with lamb′s tripe 0.2 g/kg once per day for three months; in non-administration group and negative control group, rats were gavaged with 0.9% sodium chloride solution 0.2 g/kg once per day for three months. One rat died in each for the administration group and non-administration group due to gavage. Body weight gain, pH value of gastric juice and pathological changes of gastric mucosa of the three groups were evaluated. The number of nodules on gastric mucosal surface and the incidence and scores of precancerous lesions (intraepithelial neoplasia) in gastric mucosal were analyzed. The therapeutic effects of lamb′s tripe capsule on gastric mucosal precancerous lesions in rats were evaluated. Independent sample t test , Mann Whitney U test and chi-square test were used for statistical analysis. Results:The body weight gain of rats at the 6th week in the administration group was higher than that of rats in the non-administration group ((508.26±33.96) g vs. (495.50±23.01) g), and the pH value of gastric juice of rats in the administration group was lower than that of rats in the non-administration group (3.07±0.55 vs. 4.45±0.72), and the differences were statistically significant (both P<0.05). The number of proliferative nodules on the gastric mucosal surface of the rats in the administration group was less than that of rats in the non-administration group (the ratio of gastric fundus: 6.00(3.00, 7.00) vs. 11.00(7.00, 13.00); the ratio of gastric antrum: 0.00(0.00, 1.00) vs. 3.00(2.00, 4.00); the ratio of whole stomach: 7.00(3.00, 10.00) vs. 15.00(13.00, 17.00)), and the differences were statistically significant ( U=43.50, 49.00, 49.00, all P<0.05). The score of gastric mucosal precancerous lesions in the administration group was lower than that in the non-administration group(1.00±0.00 vs.1.14±0.38), and the incidence of precancerous lesions in the administration group was lower than that in the non-administration group (1/7 vs. 5/7), and the differences were statistically significant ( t=2.45, χ2=4.67, both P=0.031). Conclusions:Lamb′s tripe capsule can significantly inhibit the progression of precancerous lesions of gastric mucosa in rats, so as to play a role in preventing the occurrence of gastric cancer.

ABSTRACT

Objective:To investigate the efficacy and related factors of lamb′s tripe extract and vitamin B12 capsule (LTEVB12) in the treatment of chronic atrophic gastritis.Methods:From October 1st 2016 to April 30th 2021, 240 patients with chronic atrophic gastritis visited the Department of Gastroenterology at Xijing Hospital, Air Force Medical University were retrospectively analyzed. All patients regularly took LTEVB12 (110 U/day, 3 times/day) for six months. At the end of treatment, endoscopy and gastric mucosal biopsy were conducted. The therapeutic effects were evaluated by comparing the changes of operative link on gastritis assessment (OLGA) and operative link on gastritis assessment based on intestinal metaplasia (OLGIM) staging before and after treatment. The related factors affecting the efficacy of the drug were analyzed. Multivariate logistic regression analysis was used for statistical analysis.Results:After half a year of treatment, the reversal efficiency of atrophy and intestinal metaplasia was 45.4% (109/240) and 37.9% (91/240), respectively, and the total efficiency was 62.9% (151/240). The reversal efficiency of OLGA and OLGIM staging reversed from high stage (stage Ⅲ to Ⅳ) to low stage (stage 0 to Ⅱ) was 53.4% (63/118) and 54.5% (36/66), respectively. The results of multivariate analysis showed that female, vitamin supplementation (≥3 times/week), negative or successful eradication of Helicobacter pylori and mild inflammatory status (inflammation score: 1 to 2) were associated with improving the efficacy of LTEVB12 (odds ratio=1.798, 3.730, 2.817 and 4.631, 95% confidence interval 1.055 to 3.064, 1.197 to 11.627, 1.171 to 6.779, 1.480 to 14.493; all P<0.05). High consumption of pickled food (≥3 times/week) was associated with reducing efficacy of LTEVB12 (odds ratio=0.384, 95%confidence interval 0.200 to 0.740). Conclusion:LTEVB12 has better reversal therapeutic effect on atrophy and intestinal metaplasia, and may reduce the risk of gastric cancer in patients with chronic atrophic gastritis.

ABSTRACT

Objective:To report two cases of steroid-resistant nephrotic syndrome (SRNS) caused by LAMB2 gene mutation, and summarize the characteristics of genotype, clinical and pathological phenotypes of children with LAMB2 gene mutation. Methods:Two cases with SRNS caused by LAMB2 gene mutation were from Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology in December 2013 and September 2019. The demographic, family history and clinical data of two cases were collected, and the peripheral blood genomic DNA was captured and sequenced by whole exome sequencing. PubMed, Medline, CNKI and Wanfang databases were searched to summarize the clinicopathological phenotypes and genotypes of patients with LAMB2 mutation. Results:Among the two cases with SRNS caused by LAMB2 gene mutation, the clinical phenotypes were all manifested as nephrotic level of proteinuria and hypoalbuminemia, and there was no extrarenal clinical manifestation. One case presented with basement membrane delamination and the other with focal segmental glomerulosclerosis (FSGS). LAMB2 mutations of two cases were Exon32 c.5390G>T(p.Cys1797Phe), Exon19 c.2557C>T(p.Arg853*) and Exon27 c.4370G>A(p.R1457Q), Exon23 c.3325G>A(p.E1109K), respectively. In literature retrieval, there were 37 cases with LAMB2 gene mutation, including 24 cases with renal biopsy data, 13 cases of focal segmental glomerulosclerosis (FSGS), 4 cases of minimal change disease, one case of diffuse mesangial sclerosis, one case of IgM nephropathy, two cases of thin basement membrane nephropathy, and three cases of mesangial hyperplasia. Among them, eight cases had basement membrane delamination tear. Among the 37 cases, 11 cases were homozygous, 22 cases were complex heterozygosity, and 4 cases were heterozygous mutation. Conclusions:LAMB2 mutation may cause delamination tear of glomerular basement membrane. The clinical phenotype is congenital nephrotic syndrome or SRNS. The literature review shows the extrarenal manifestations caused by LAMB2 mutation are mostly various ocular abnormalities, as well as respiratory, digestive and nervous system abnormalities, and the time of progression to end-stage renal disease is also different.

Objective To investigate the main chemical constituents of the low polarity extracts from pinusmassoniana Lamb. leaves and their synergetic activity with fluconazole against fluconazole-resistant Candida albicans. Methods The pinusmassoniana leaves were extracted with 80% ethanol, and then the extracts were extracted by petroleum ether to obtain the low polarity extracts. The chemical components were detected by GC-MS and elucidated by the comparison with the standard mass spectral data. The relative contents in percentage were calculated using the area normalization method. The minimal inhibitory concentrations (MIC80) of fluconazole-resistant Candida albicans strains by the low polarity extracts in combination with fluconazole were determined by checkerboard microdilution assay. Results 30 components were detected from the low polarity extracts, and 17 components were identified. The minimum inhibitory concentration (MIC80) of the 80% ethanol extracts, the low polarity extracts and the petroleum ether extracts from the pinusmassoniana leaves combined with fluconazole against fluconazole-resistant Candida albicans were 7.81 μg/ml, 31.25 μg/ml and >250 μg/ml, respectively. Conclusion The 80% ethanol extracts of pinusmassoniana leaves and its low polarity extracts have synergistic activity combined with fluconazole onfluconazole-resistant Candida albicans. The diterpenoids (53.99%) may be the effective components of the low polarity extracts.

ABSTRACT: The aim of this study was to evaluate the influence of different non-genetic effects on indicator traits for maternal ability in Santa Inês ewes. Data included performance records of 100 lambs (males and females) born from 59 dams, from 2009 to 2012. The analyzed traits were birth weight (BW), weaning weight (WW), average daily gain from birth until weaning (ADGBW), total litter weight at birth (TLWB), and total litter weight at weaning (TLWW). The effects analyzed were the year of birth of the lamb, birth season, dam age at lambing, dam weight at lambing, sex of the lamb, lamb birth type, interaction between sex and birth type, and interaction between sex and birth season. SAS® software (SAS University Edition, USA) was used for calculation of the analysis of variance, means, and Pearson correlation coefficients. With the exception of the birth season, all the other environmental effects evaluated had a significant influence on at least one of the studied traits. The correlation estimates ranged from low to high and were either positive or negative. Birth weight was negatively correlated with the birth type and influenced positively all the other performance traits evaluated. The maternal ability of Santa Inês ewes was more clearly influenced by the age and weight of the dam at lambing, and the lamb birth type.


RESUMO: O objetivo deste estudo foi avaliar a influência de diferentes efeitos não-genéticos sobre características indicadoras de habilidade materna em ovelhas da raça Santa Inês. Os dados utilizados incluíram registros de desempenho de 100 cordeiros (machos e fêmeas) filhos de 59 ovelhas, nascidos de 2009 a 2012. As características analisadas foram peso ao nascimento (PN), peso ao desmame (PD), ganho médio diário do nascimento ao desmame (GMND), peso total das crias ao nascer (PTCN) e peso total das crias ao desmame (PTCD). Os efeitos analisados foram ano de nascimento da cria, estação de nascimento, idade da mãe ao parto, peso da mãe ao parto, sexo da cria, tipo de nascimento da cria, interação entre sexo e tipo de nascimento, e interação entre sexo e estação de nascimento. O programa SAS (SAS University Edition, EUA) foi utilizado para o cálculo de análise de variância, médias e coeficientes de correlação de Pearson. Com exceção da estação de nascimento, os demais efeitos avaliados exerceram influência significativa sobre pelo menos uma das características estudadas. As estimativas de correlação variaram de baixa a alta magnitude e foram tanto positivas quanto negativas. O peso da cria ao nascimento foi negativamente correlacionado com o tipo de nascimento e influenciou positivamente todas as outras características de desempenho avaliadas. A habilidade materna de ovelhas Santa Inês foi mais claramente influenciada pela idade e peso da mãe ao parto e pelo tipo de nascimento da cria.

ABSTRACT

The aim of this study was to evaluate the lung maturity of premature and full-term lambs by analyzing amniotic fluid using the following methods: Clements test, Nile blue cytology test, hematoxylin-Shorr stain, lamellar body count, and radiographic tests. The use of these methods is intended to identify high-risk newborns and provide immediate clinical intervention after birth. Altogether, 56 animals (24 ewes and 32 lambs) were included in the study and divided into 3 groups. Group I consisted of 8 ewes that were at approximately 145 days of gestation; this group delivered 10 lambs naturally. Group II consisted of 8 ewes that were at 138 days' gestation; this group delivered 11 lambs by cesarean section. Group III consisted of 8 ewes at 138 days' gestation; this group was administered intramuscular dexamethasone (16mg/animal) 36 hours prior to a cesarean section. Group III delivered11 lambs. Cytological tests were performed using a microscope with a maximum magnification of 1000x, while the Clements test was visually observed by one of the researchers. Amnioticfluid lamellar body counts were measured using transmission electron microscopy. Among the staining methods, hematoxylin-Shorr was reliable, and Group III had a greater number of orangeophilic cells when compared to Group II, probably due to corticoid administration. The Clements test showed pulmonary maturity in approximately 20% of Group I lambs and Group II showed 9.1% of bubbles; however, Group III had the highest pulmonary maturity percentage (36.4%). The lamellar bodies were measured, and all groups had sizes between 0.019 and 0.590μm. Radiographic evaluation revealed that the majority of lambs presented some level of pulmonary radiodensity, indicating an acinar pattern at birth. These results are in line with the expectations of each group. We found that the normal group showed greater pulmonary maturity, whereas Group II presented pulmonary immaturity, which is expected because this group comprised lambs born prematurely and Group III showed pulmonary maturity almost comparable to the normal delivery group (Group I). This is due to the fact that although these animals are premature, the use of dexamethasone helped in pulmonary maturation. Therefore, these pulmonary maturity tests are considered effective when more than one technique is used and can be used routinely in the care of a pregnant ewe in labor, where a simple collection of amniotic fluid can predict a high-risk pregnancy and alert the veterinarian if the newborn needs intensive supportive treatment.(AU)


O objetivo deste estudo foi avaliar a maturidade pulmonar de cordeiros prematuros e a termo por meio da análise do líquido amniótico utilizando os seguintes métodos: teste de Clements, teste de citologia do azul do Nilo, coloração de hematoxilina-Shorr, contagem de corpos lamelares e testes radiográficos. Um desses métodos tem por objetivo identificar recém-nascidos de alto risco e fornecer intervenção clínica imediata após o nascimento. Ao todo, 56 animais (24 ovelhas e 32 cordeiros) foram incluídos no estudo e divididos em 3 grupos. O grupo I foi composto por 8 ovelhas com aproximadamente 145 dias de gestação; este grupo deu à luz 10 cordeiros naturalmente. O Grupo II foi composto por 8 ovelhas com 138 dias de gestação; este grupo deu à luz 11 cordeiros por cesariana. Grupo III consistiu de 8 ovelhas com 138 dias de gestação; este grupo recebeu dexametasona intramuscular (16 mg / animal) 36 horas antes de uma cesariana. O Grupo III entregou 11 cordeiros. Os testes citológicos foram realizados em microscópio com aumento máximo de 1000x, enquanto o teste de Clements foi observado visualmente por um dos pesquisadores. A contagem de corpos lamelares de líquido amniótico foi medida usando microscopia eletrônica de transmissão. Dentre os métodos de coloração, o hematoxilina-Shorr foi confiável, sendo que o Grupo III apresentou maior número de células orangeofílicas quando comparado ao grupo II, provavelmente devido à administração de corticóide. O teste de Clements mostrou maturidade pulmonar em aproximadamente 20% dos cordeiros do Grupo I e o Grupo II apresentou 9,1% de bolhas; entretanto, o Grupo III apresentou o maior percentual de maturidade pulmonar (36,4%). Os corpos lamelares foram medidos e todos os grupos apresentaram tamanhos entre 0,019 e 0,590μm. A avaliação radiográfica revelou que a maioria dos cordeiros apresentava algum grau de radiodensidade pulmonar, indicando padrão acinar ao nascimento. Esses resultados estão alinhados com as expectativas de cada grupo. Verificamos que o grupo normal apresentou maior maturidade pulmonar, enquanto o Grupo II apresentou imaturidade pulmonar, o que é esperado por se tratar de cordeiros nascidos prematuramente e o Grupo III apresentou maturidade pulmonar quase comparável ao grupo de parto normal (Grupo I). Isso se deve ao fato de que, embora esses animais sejam prematuros, o uso da dexametasona auxiliou na maturação pulmonar. Portanto, esses testes de maturidade pulmonar são considerados eficazes quando mais de uma técnica são utilizadas e podem ser usadas rotineiramente no cuidado de uma ovelha gestante em trabalho de parto, onde uma simples coleta de líquido amniótico pode prever uma gravidez de alto risco e alertar o veterinário se o recém-nascido precisa de tratamento de suporte intensivo.(AU)

ABSTRACT

Haemonchus contortus is the major gastrointestinal parasite of sheep raised in tropical and subtropical areas worldwide. This trial aimed to evaluate the influence of H. contortus infection on the bone marrow response of Santa Ines (SI) and Ile de France (IF) suckling lambs experimentally infected with H. contortus. Fourteen SI lambs and 12 IF lambs were randomized in four groups: infected SI (n=8), non-infected SI (n=6), infected IF (n=8) and non-infected IF (n=4). Lambs of infected groups were submitted to 27 infections, conducted every two days, from 14 to 68 days of age, and each lamb received a total of 5400 H. contortus infective larvae (L3). Ten blood samples were obtained during the experimental period to be used for erythrocyte and leukocyte counts, packed cell volume and total plasma protein estimation. Additionally, it was carried out a differential leukocyte count. Lambs from control groups did not shed eggs in faeces all over the experiment, while infected Santa Ines and Ile de France lambs presented means of 2963 EPG (Eggs Per Gram of faeces) and 8175 EPG in the last sampling (P<0.05), respectively. Infected Santa Ines lambs had an increase in eosinophil release, however differences (P<0.05) on circulation number in comparison with infected Ile de France lambs were identified only in the last sampling (54 days post first infection). The mild H. cortortus infection did not produce significant changes in the blood variables of the Ile de France and Santa Ines suckling lambs.(AU)


Haemonchus contortus é o principal parasita gastrintestinal de ovinos criados em áreas tropicais e subtropicais. Este estudo objetivou avaliar a influência da infecção por H. contortus na resposta medular de cordeiros lactentes das raças Santa Inês (SI) e Ile de France (IF) infectados experimentalmente com H. contortus. Quatorze cordeiros SI e 12 cordeiros IF, foram distribuídos em quatro grupos: SI infectado (n=8), SI não infectado (n=6), IF infectado (n=8) e IF não infectado (n=4). Cordeiros dos grupos infectados foram submetidos a 27 infecções, realizadas a cada dois dias, do 14º até 68º dia de vida, com um total de 5400 larvas infectantes (L3) de H. contortus por animal. Foram realizadas 10 coletas de sangue ao longo do período experimental para determinação do volume globular, proteínas plasmáticas totais, contagem de eritrócitos e leucócitos, além do diferencial de leucócitos. Cordeiros controles não infectados não eliminaram ovos nas fezes durante todo o experimento, enquanto que os cordeiros Santa Ines e Ile de France infectados apresentaram em média 2963 ovos por grama de fezes (OPG) e 8175 OPG na última coleta (P<0,05), respectivamente. Cordeiros Santa Inês infectados tiveram um aumento na produção de eosinófilos, mas diferenças (P<0.05) no número de eosinófilos sanguíneos em comparação a cordeiros Ile de France infectados foram detectadas somente na última coleta (54 dias após a primeira infecção). A infecção leve por H. cortortus não induziu alterações significativas nas variáveis sanguíneas de cordeiros lactentes Ile de France e Santa Inês.(AU)

Abstract Background: Carcass and sheep meat quality are influenced by several factors, such as race, age, slaughter weight, type of diet, and dietary supplementation. Objective: To evaluate the effect of castration and vitamin E supplementation on performance, carcass characteristics and meat of lambs slaughtered at 45 kg. Methods: Thirty-two male lambs (16 castrated and 16 not castrated) were fed diets with or without the inclusion of vitamin E. The animals had an average age of 180 ± 20 days and a mean weight of 25.1 ± 4.6 kg. Performance was evaluated for 56 days, and the animals were slaughtered when reaching a weight of 45 kg. The experimental design was randomized blocks based on initial weights, and variables were assessed by a 2x2 factorial design (castrated and intact, supplemented or not with vitamin E, with a probability of 5%). Results: Higher proportion (p<0.05) of visceral fat was found in castrated animals (kidney: 2.45%, inguinal: 0.57% and omental: 4.96%) than in intact animals (kidney: 1.25%, inguinal 0.27%. and omental 3.12%). The aged meat samples showed no color differences for any of the treatments, but there were differences in shear force for steaks aged for 7 days, where the castrated animals showed an average of 2.64 kgf and intact animals showed an average 3.29 kgf. The animals supplemented with vitamin E had lower shear forces in non-aged samples. Conclusion: Castration increased deposition of visceral and fat coverage but did not influence the cut yields and total carcass yield. The animals supplemented with vitamin E had lower shear forces in non-aged samples.


Resumen Antecedentes: La calidad de la canal y de la carne de ovino puede ser influenciada por varios factores, como raza, edad, peso de sacrificio, tipo de dieta y suplementación. Objetivo: Evaluar el efecto de la castración y de la suplementación con vitamina E sobre el desempeño, la calidad de la canal y la carne de corderos Santa Inés sacrificados a los 45 kg de peso. Métodos: Fueron utilizados treinta y dos corderos: dieciséis castrados y dieciséis no castrados, alimentados con dietas con o sin inclusión de vitamina E. Los animales tenían una edad promedio de 180 ± 20 días y un peso promedio de 25,1 ± 4,6 kg. Fue evaluado el desempeño durante 56 días y, al llegar a los 45 kg de peso fueron sacrificados. El diseño experimental fue en bloques aleatorizados con base en el peso inicial, y las variables analizadas mediante un modelo factorial 2x2 (castrado o no castrado y suplementado o no con vitamina E, con una probabilidad del 5%). Resultado: Se encontró mayor proporción (p<0,05) de grasa visceral en los animales castrados (renal: 2,45%; inguinal: 0,57% y mesentérica: 4,96%) que en los no castrados (renal: 1,25%; inguinal 0,27% y mesentérica: 3,12%). Las muestras de carne maduradas no presentaron diferencias de color entre tratamientos, sin embargo hubo diferencias en la fuerza de corte para las carnes maduradas por 7 días, donde los animales castrados presentaron una media de 2,64 kgf y los no castrados de 3,29 kgf. La carne de los animales suplementados con vitamina E tuvo menor fuerza de corte en las muestras no maduradas. Conclusión: La castración aumentó la deposición de grasa visceral y de cobertura pero no influenció las características de rendimiento total de la canal y cortes. Los animales suplementados con vitamina E tuvieron menor fuerza de corte en las muestras no maduradas.

Métodos: Foram utilizados trinta e dois cordeiros, dezesseis castrados e dezesseis não castrados alimentados com dietas com ou sem inclusão de vitamina E. Os animais tinham idade média de 180 ± 20 dias e peso médio de 25,1 ± 4,6 kg. Foi avaliado o desempenho durante 56 dias, e ao chegarem aos 45 kg foi realizado o abate. O delineamento experimental foi em blocos ao acaso, com base no peso inicial, e as variáveis analisadas por modelo fatorial 2x2 (castrado ou não castrado e suplementado ou não com vitamina E, ao nível de 5%). Resultado: Foi encontrada maior proporção (p<0,05) das gorduras viscerais nos animais castrados (renal: 2,45%; inguinal: 0,57% e omental: 4,96%) do que nos animais não castrados (renal: 1,25%; inguinal 0,27% e omental: 3,12%). As amostras maturadas não apresentaram diferenças de cor entre os tratamentos, no entanto houve diferenças na força de cisalhamento para as carnes maturadas por 7 dias, na qual os animais castrados apresentaram média de 2,64 kgf e os não castrados de 3,29 kgf. Os animais suplementados com vitamina E apresentaram menores forças de cisalhamento nas amostras não maturadas. ABSTRACT: This study described the pathological aspects of a mature intracranial teratoma with meningocele in a one-day-old lamb. The diagnosis of mature teratoma was based on microscopic features, where well-differentiated tissues representative of the three germ layers were observed. The tumor consisted of respiratory, gastrointestinal and urinary epithelium as components of the endoderm; muscle and bone tissues as components of the mesoderm; and keratinized stratified squamous epithelium and adnexa as components of the ectoderm. To the best of our knowledge, this is the first report of intracranial teratoma with meningocele in animals.


RESUMO: Descrevem-se os aspectos patológicos de um teratoma intracraniano benigno com meningocele em um cordeiro de um dia de idade. O diagnóstico de teratoma benigno foi baseado nos achados histopatológicos, com tecidos representativos das três camadas germinativas. O tumor consistia de epitélio respiratório, gastrintestinal e urinário como componentes do endoderma; tecido muscular e ósseo do mesoderma; e epitélio escamoso estratificado queratinizado e anexos como componentes do ectoderma. Com base no conhecimento dos autores, este é o primeiro relato de teratoma intracraniano com meningocele em animais.

ABSTRACT

The study evaluate the frequency of poisoning by Brachiaria spp. at the rearing, growth and termination stages in various lamb breeds at increasing supplementation levels. Forty-five lambs were used in the growth phase in pastures of Brachiaria spp. with a history of having induced poisoning. The lambs were distributed in nutritional treatments: lambs receiving mineral supplementation (MS), energy/protein supplementation (EPS) at 0.8% of body weight (bw), EPS at 1.6% bw and EPS at 2.4% bw. The lambs were allotted two flocks (F1 and F2) of 21 and 24 lambs each. Clinical signs of poisoning were observed in all treatments. All MS lambs died. The frequency of poisoning were highest in the 0.8% EPS and 1.6% EPS treatments. All lambs in the 2.4% EPS treatment recovered. F1 lambs had a higher frequency of poisoning than F2 lambs regardless of nutritional treatment. Morbidity rates for the F1 and F2 lambs were 52.3 and 16.7%, respectively. Supplementation was not sufficient to decrease the frequency of poisoning in lambs at the termination stage, which was dependent on the genetic origin of the lambs. EPS of 2.4% bw treatment, was an efficient nutritional strategy to minimize the effects of poisoning in lambs fed on Brachiaria spp.(AU)


O objetivo deste estudo foi avaliar a frequência da intoxicação por Brachiaria spp. nas fases de recria, crescimento e terminação em várias raças de cordeiros em níveis crescentes de suplementação. Quarenta e cinco cordeiros foram utilizados na fase de crescimento em pastagens de Brachiaria spp. com história de intoxicação, distribuídos em diferents tratamentos nutricionais: cordeiros recebendo suplementação mineral (SM), suplementação energética/protéica (SEP) a 0,8% do peso vivo (pv), SEP a 1,6% pv e SEP a 2,4% pv. Vinte e um cordeiros vieram do lote F1 e 24 cordeiros foram do lote F2. Sinais clínicos de intoxicação foram observados em todos os tratamentos. Todos os cordeiros com MS morreram. As frequências de intoxicação foram maiores nos tratamentos com 0,8% e 1,6% de SEP. Todos os cordeiros no tratamento com 2,4% de SEP se recuperaram. Cordeiros F1 apresentaram maior frequência de intoxicação que os cordeiros F2, independentemente do tratamento nutricional. As taxas de morbidade para os cordeiros F1 e F2 foram de 52,3 e 16,7%, respectivamente. A suplementação não foi suficiente para diminuir a freqüência de intoxicação em cordeiros na fase de terminação, dependente da origem genética dos cordeiros. O SEP de 2,4% pv, no entanto, foi eficiente para minimizar os efeitos da intoxicação em cordeiros alimentados com Brachiaria spp.(AU)

Abstract This article contains a morphological description of a case of early presentation lymphoma in a lamb, characterised by its unusual clinical history and advanced lesions. The primary tumour was located in the skin in the carpal region, and metastases were observed in the brain, right eye, all the pulmonary lobes, the myocardium and endocardium of the heart, left adrenal gland, abdominal portion of the vagal nerve, serosa of the rumen, the body of lumbar vertebra L3, and the submandibular, mediastinal and mesenteric lymph nodes. Histopathological examination revealed that it was a large cell lymphoma. Immunohistochemical analysis was carried out with antibodies for MUM1/IRF4, CD79a, CD3, CD10 and Ki67, used here for the first time as markers to characterise a lymphoma in this species. The result revealed that it was a B-cell lymphoma.


Resumen En este artículo se describe morfológicamente un caso de linfoma de presentación temprana en la especie ovina, el cual se caracterizó por lo inusual de su historia clínica y lo avanzado de las lesiones. El tumor primario se localizó en la piel del carpo y se observaron metástasis en el cerebro, el ojo derecho, todos los lóbulos pulmonares, el miocardio y endocardio del corazón, la glándula adrenal izquierda, el nervio vago en su porción abdominal, la serosa del rumen, el cuerpo vertebral lumbar L3, los linfonodos submandibulares, mediastínicos y mesentéricos. El examen histopatológico reveló un linfoma no Hodgkin de células grandes. El análisis inmunohistoquímico se realizó con anticuerpos para MUM-1/IRF-4, CD79a, CD3, CD10 y Ki-67, los cuales se usaron por primera vez como marcadores para caracterizar un linfoma en esta especie. El resultado reveló que se trató de un linfoma de células-B.


Resumo Neste artigo descreve-se morfologicamente um caso de linfoma de apresentação precoce na espécie ovina, caracterizado pela historia clínica incomum e as lesões avançadas. O tumor primário foi encontrado na pele do carpo e metástase foi observada no cérebro, olho direito, todos os lobos pulmonares, miocárdio e endocárdio do coração, a glándula adrenal esquerda, nervo vago na porção abdominal, a serosa do rúmen, o corpo vertebral lombar L3, linfonodos submandibulares, mediastinos e mesentéricos. O exame histopatológico revelou linfoma não-Hodgkin de células grandes. A análise imuno-histoquímica foi realizada com anticorpos para MUM-1/IRF-4, CD79a, CD3, CD10 e Ki-67, os quais usaram-se por primeira vez como marcadores para caracterizar um linfoma nessa espécie. O resultado revelou que se tratou de linfoma de células-B.

Laminins are a major constituent of the extracellular matrix (ECM). Laminin-111, the most extensively studied lamininisoform, consists of the a1, the b1 and the c1 chain, and is involved in many cellular processes, like adhesion, migrationand differentiation. Given the regulatory role of phosphorylation in protein function, it is important to identify thephosphorylation sites of human laminin b1-chain sequence (LAMB1). Therefore, we computationally predicted all possiblephosphorylation sites in LAMB1. For the first time, we identified the possibly responsible kinases foralready in vitro experimentally observed phosphorylated residues in LAMB1. All known functional (active) sites ofLAMB1, were recorded after an extensive literature search and combined with the experimentally observed and ourpredicted phosphorylated residues. This generated a detailed phosphorylation map of LAMB1. Five kinases (PKA, PKC,CKII, CKI and GPCR1) were indicated important, while the role of PKA, PKC and CKII, kinases known for ectophosphorylation activity, was highlighted. The activity of PKA and PKC was associated with the active site RIQNLLKITNLRIKFVKLHTLGDNLLDS. Also, predicted phosphorylations inside two amyloidogenic (DSITKYFQMSLE,VILQHSAADIAR) and two anti-cancerous (YIGSR and PDSGR) sites suggested a possible role in the development of thecorresponding diseases.

ABSTRACT: The objective of this study was to better understand farmers' perceptions regarding sheep welfare and procedures concerning the tail docking of sheep in the state of Parana, Southern Brazil. The study was carried out via telephone interviews or personally with 146 sheep farmers. Twenty-eight farmers (19.2%) did not tail dock; the main reasons given were because they raised short-hair sheep breeds. One hundred and eighteen farmers docked their sheep tails (80.8%). The main reasons given were hygiene (61.0%), facilitated mating (42.4%), breed standards (29.7%), and esthetics (26.3%). Rubber ring was the main method used for tail docking. Although farmers in Parana recognized that sheep are sentient animals and that tail docking causes pain, anesthesia was used by only six (5.1%) farmers during the procedure of tail docking. Our results showed that farmers' opinions on sheep tail docking are divergent. Farmers who perform tail docking must be encouraged to use anesthesia and analgesia, and to ponder on the real need for the practice. This statement seems especially relevant considering the fact that some farmers reported the viability of maintaining wool sheep with undocked tails and that they recognized the suffering involved in this procedure.


RESUMO: O objetivo deste trabalho foi compreender melhor a percepção de ovinocultores em relação ao bem-estar de ovinos e aos procedimentos envolvendo a caudectomia desses animais no Paraná, sul do Brasil. O estudo foi realizado por meio de entrevistas telefônicas ou pessoalmente com 146 ovinocultores. Vinte e oito ovinocultores (19,2%) não realizavam caudectomia; a principal razão citada foi porque os produtores criavam ovelhas deslanadas (pelo curto). Cento e dezoito ovinocultores realizavam a caudectomia (80,8%) e as principais razões citadas foram: higiene (61,0%), monta facilitada (42,4%), padrão racial (29,7%) e estética (26,3%). O principal método utilizado para realizar a caudectomia foi o anel de borracha. Apesar dos ovinocultores no Paraná reconhecerem que ovinos são animais sencientes e que a caudectomia causa dor, somente seis (5,1%) aplicavam anestesia durante o procedimento. Nossos resultados demonstram que a opinião dos ovinocultores com relação à caudectomia é divergente. Ovinocultores que realizam a caudectomia devem ser encorajados a usar anestesia e analgesia, além de ponderar a real necessidade para essa prática. Essa afirmação se mostra relevante ao considerar o fato de que alguns produtores relataram a viabilidade de manter ovinos lanados com cauda inteira e o reconhecimento por parte deles do sofrimento causado aos animais durante a remoção da cauda.

Objective To establish a method to determine the contents of total flavonoids and shikimic acid in pine needles of Pinus massoniana Lamb.in Wudang Area.Methods Rutin was used as reference standard,and the content of total flavonoids in pine needles of Pinus massoniana Lamb. was determined by UV spectrometry at wavelength of 500 nm. The content of shikimic acid was determined by HPLC-DAD. The Fortis Xi C8 column (5 μm, 250 mm × 4.6 mm) was adopted with acetonitrile - 0.4% phosphoric acid solution (8:92, V/V) as mobile phase at the flow rate of 0.9 mL/min. The detection wavelength was 213 nm; the column temperature was 30 ℃; the injection volume was 20 μL. Results The linear range was 8.26-49.54 μg for rutin (r=0.999 4) with an average recovery of 99.2%, RSD=1.94%. The linear range was 10.26-61.56 μg for shikimic acid (r=0.999 4) with an average recovery of 99.5%,RSD=1.93%.The contents of total flavonoids in pine needles of Pinus massoniana Lamb.was 28.33 mg/g, and shikimic acid was 15.25 mg/g, respectively. Conclusion The method is simple, rapid, accurate and reproducible. It can be used for the content determination of total flavonoids and shikimic acid in pine needles of Pinus massoniana Lamb. in Wudang Area.
